Activation of inflammation and endothelin has been reported to be the cause of acidosis related kidney injury. However, the role and change of intrarenal renin-angiotensin system (RAS) during metabolic acidosis are uncertain. The present study aimed to investigate intrarenal RAS and inflammation in healthy rats with excessive acid loading. Methods: Male Sprague-Dawley rats were fed with water containing 0.14M NH4Cl to induce metabolic acidosis. Daily urine was collected for biochemical analysis and the animals were harvested for blood sampling and renal tissue analysis at 1 and 8 weeks intervals. By using RT-PCR, RAS components including renin, rennin/prorenin receptor, angiotensinogen, angiotensin converting enzyme (ACE), angiotesin II type 1 and 2 receptor (AT1R/AT2R) were analyzed. The expression of tranforming growth factor- (TGF-) and nuclear factor-kappa B (NFk-B) were also assessed to evaluate inflammatory status. Protein abundance of the molecules was analyzed by western-blotting. Results: Acidemia with bicarbonate concentration was found in 1 week but not 8 -week loaded rats. An increase in daily proteinuria and decline of glomerular filtration rate (GFR) were observed in acid loaded rats. In 1-week acidosis group, angiotensinogen (359.6 62.4% of control), ACE (577.8 107.3%), AT1R (194.1 25.2%) and AT2R (4629.4 1525.5%) were significantly increased (all p0.05). Persistent increase of AT1R expression (168.1 12.7% of control, p0.05) was found as NH4Cl was continued for 8 weeks. There was no significant changes in TGF- and NF-B of both 1-week and 8-week animals.
