Bcl—2、NF—KB在腮腺腺样囊性癌中表达和临床意义

1、bel2.nfkb在腮腺腺样囊性癌中表达和 临床意义摘要目的:探讨bel-2和nf-kb在腮腺腺样囊性癌中的表达及意义。方法：应用免疫组化sp法检侧49例 腮腺腺样囊性癌和20例正常腮腺组织中bel-2和nf-kb的 表达情况，统计学分析采用x2检验，p0. 05）,与tnm分期 有关，bel-2和nf-kb两者存在正相关性（p0. 05）（见表 2）o bel-2在tnm临床分期的i、ii期阳性表达率85.71%明显高于111.iv期的53. 57%,1、11期中、强阳性表达率76. 19%也明显高于111、iv期的25. 0%,研究结果显示bel-2总阳性表达率和表达强度随着肿瘤tnm分期

2、的增加呈现下降趋 势，be 1-2在tnm临床分期的i、ii期和111、iv期经统计 学比较有显著性差异（p2. 2 nf-kb免疫组化结果：49例腮腺腺样囊性癌标本中34例nf-kb出现阳性染色，染色 主要定位于癌细胞胞浆，少部分定位于细胞核，染色较均匀;20例正常腮腺组织中腺泡全部阴性染色，有7例导管系统出现弱阳性染色，阳性染色部位定位于管腔上皮细胞胞浆中，染色分布均匀，统计学分析两组差别有显著性差异（p0.05）（见表5）。nf-kb在tnm临床分期的i、ii期阳性表达率明显髙于111、iv期，i、ii期中、强阳性表达率也明显高于111、iv期，nf-kb在tnm临床分期的i、ii期和1

3、11、iv 期经统计学分析有显著性差异（p 现已证明bcl-2的 调节位点上含有nf-kb的结合位点13 o tamatani20等也 证实阻断nf-kb信号传导通路可减少下游抗凋亡蛋白bcl-2 的表达。nf-kb与bcl-2在腮腺腺样囊性癌抗凋亡相关性的 研究国外报道罕见，国内未见报道。本研究发现bel- 2和nf-kb在腮腺腺样囊性癌的发生、发展过程中密切相关，且两者之间存在正相关关系。推测在腮腺腺样囊性癌发生和发 展过程中刺激因素先激活nf-kb信号传导通路，nf-kb继而使下游抑制细胞凋亡基因bcl2表达增强，bcl2通过使癌 细胞长期处于g1期而延长了其增殖周期，进而发挥抑制细 胞
