7、prepared by suspending colonies into Mueller-Hinton broth (CAMHB) (containingCiprofloxacin hydrochloride) from 18 to 24 h (B. anthracis) or 42 to 48 h (Y. pestis) on sheep blood agar(SBA) plates that are incubated at 35C. Suspended cultures are diluted with CAMHB to a bacterial celldensity of 105 CF

8、U/mL adjusted based on the optical density at 600 nm. To each well of the 96-well plate, 50L of dilutions is added for a final inoculum of 5104 CFU/well. Plates are incubated at 35C. MICs aredetermined visually at 18 to 24 h (B. anthracis) or 42 to 48 h (Y. pestis) and also by absorbance at 600 nm2.

The mice (n=3/time point/group) are culled at 1, 10, 20, or 30 min and 1, 1.5, 2, 4, 8, 12 h followingCiprofloxacin hydrochloride (Bay-09867 (hydrochloride) administration and 1, 15, or 30 min and 1, 2, 4, 6,10, 18, or 24 h following DRCFI or CFI administration. Blo

11、od sampling points are chosen based upon theshort half-life of Ciprofloxacin hydrochloride and longer half-life of CFI. Blood and lungs (whole organ) arecollected post mortem for analysis. The lung doses following CFI or DRCFI administration are calculatedusing the concentration of Ciprofloxacin hyd
