Theurgencyforchangeisclear.Ouranalysisof

FDACompleteResponseLetters(CRLs)issuedto

toppharmaceuticalcompaniesbetween2009

and2024showsthatlastyearwasunprecedented:

overhalf(64%)ofmarketdelayswereduetochemistry,manufacturingandcontrols(CMC)issues,delayingbothpatientaccessandrevenues.3Mostofthese

setbacksstemmedfromtheproductionofcomplexbiologics,underscoringthestrainontoday’s

technicaloperationsfunctions.

Forexecutives,thestakescouldnotbehigher.Ourresearchfoundthatcompaniesmodernizingtheirmanufacturingoperationsstandtocapturethreecriticaladvantages:

•Secureanuninterruptedsupplyofmedicinestopatients.

Thecurrentstate:Progressamidpersistentchallenges

Ourresearchindicatesthatthepharmaceuticalsectorisatapivotaljunctureinitsdigitaltransformationjourney.Anewwaveofadvancedanalyticsand

AIisrapidlyemerging,yetadoptionvariessignificantlyacrossorganizations.

Inourglobalsurvey,5asignificantportionoftopbiopharmaorganizations

remain“stuckinthemiddle”oftheirdigitaljourney.35%ofsurveyedexecutivesindicatedtheircompaniesfunctionas“connectedorganizations.”Ourfollow-upinterviewsrevealedthatevenamongthoseachievinghighlevelsofadvanced

intelligenttechnologiesadoption,significantdisparitiespersistacrossbusinessunits,modalitiesandsites.19%oftopbiopharmaexecutivesreportedoperatingina“predictive”environment.6Thismeanstheyareleveragingintegrated

systemsthatcombinehistoricalandreal-timedatawithAImodelstoenable

proactiveprocessoptimization,enhanceoperationalefficiencyandsupport

data-drivendecision-making.Lessthan10%ofrespondentsreportedoperatinginan“adaptive”environment,havingpartiallyautonomous,self-optimizing

processespoweredbyAI,machinelearning(ML)androbotics.

andleanercoststructures.Anadvancedexample,sharedbyanexecutivefromaleadingglobal

biopharmacompany,involvedembeddingAIandreal-timeanalyticsintoitsmanufacturingoperations.Thisapproachwasusedtoaddressvariabilityintheirkeybiologicproduct.Bycontinuouslyanalyzing

thousandsofreal-timeprocessvariablesusing

machinelearning,thesystemidentified10–15keypredictorsthatmoststronglyinfluencedyield.

Thisenabledoperatorstointerveneproactively—withinvalidatedranges—beforedeviationscouldimpactoutcomes.

Asaresult,thecompanyhascreatedasmarter,moreadaptiveproductionenvironmentthat

ensureshigherefficiencyandproductquality,

reducingwasteinmanufacturing.AllofthiswaspossiblebecauseofembeddingAIdirectlyintolivemanufacturingoperations.

Similarly,45%ofexecutiveshighlightedmajoradvancementsintechnologytransfer.

Onecompanyimplementedcomputer-based

simulationmodeling(knownasinsilicomodeling)topredictoptimalmanufacturingparameters

priortophysicalexecution.Thisapproachprovedespeciallyvaluableduringthetransferofan

acquiredassetwithlimitedpriordocumentation.Asaresult,theyachievedtheirfastesttech

transferofanassetthroughacquisitiontodate.

Theroadblocksonthewaytoscaledigitalinitiatives

Whiletherearegreatsuccessstories,mostcompaniestodaystandat

aninflectionpoint(seeFigure2).Thosethatsucceedinindustrializing

theirdigitalcapabilitieshavetheopportunitytoscalerapidlyandprogresstowardfullyautonomous,end-to-endoperations.7

Yet,thispotentialremainslargelyuntapped.Ourresearchrevealsthreepersistentbarriersholdingbackprogress:

•Digitalpilotsareoftenconfinedtolow-risk,low-impactareas.

•Coredatainfrastructureandsystemsarenotyetreadytosupportscale.

•Initiativesremainsiloedwithinfunctions,eg.productdevelopment,MS&T,regulatoryandqualityandIT/digital.

Theresult?Fragmentedsolutionswithlimitedreturnoninvestment(ROI),digitalinitiativesthatcannotscaleandagrowingnumberof“digitaldeadends”.8Thesedisconnectsarewideningthegapbetweenambitionand

execution.Ifleftunaddressed,theindustry’sdigitalmomentumcouldstall,orworserequirecostlyrework.
