( 1 ) immobilized microbial membrane be the yeast trichosporon cutaneum, ifo-10466 sealed in a membrane under a state wherein its physiological function is kept. an immobilized microbial membrane shall (2) oxygen electrode measurement of dissolved oxygen specified in 7.3 (1) (electrode) of jis k 0803. an oxygen electrode shall be an electrode for (3) microbial sensor a microbial sensor shall be constructed by combining an immobilized microbial membrane with an electrode which detects the variation of the substance concentration to be generated when its microbe has decomposed and ingested organic substance, for example, the decrease of oxygen concentration following the increase of the respiration activity of microbe. 3. structure of microbial sensor and measuring system 3.1 immobilized microbial membrane an immobilized microbial membrane shall be of the type which covers the surface of an oxygen electrode. 3.2 oxygen electrode either of the following electrodes. the oxygen electrode shall be in accordance with ( 1) diaphragm polarograph type oxygen electrode (2) diaphragm galvanic cell type oxygen electrode 3.3 measuring method of bods value the measuring method of a bods value shall be in accordance with -either of the following systems. (1) a system of calculation from the maximum variation amount of oxygen electrode current. (2) a system of calculation from the variation amount after a specific time of oxygen electrode current. 4 . rated voltage and rated frequency the rated voltage of an apparatus for estimation shall be single phase ac 100 v and the rated frequency thereof shall be 50 hz, 60 hz, or common 50 hz/60 hz. 5, performance an apparatus for estimation shall conform to the following performances. (1) repeatability the deviation shall be f 10 % of the mean value. when a test is performed by the testing method of 7.3 ( i ) , (2) response time (2), the response time shall be within 20 min. when a test is performed by the testing method of 7.3 (3) stability to voltage fluctuation method of 7.3 (3), the variation of its indication value shall be 4 3 %. when a test is performed by the testing (4) insulation resistance 7.3 (4), its value shall be 5 ms1 or over. when a test is performed by the testing method of (5) withstand voltage (5), any abnormality shall not be generated, when a test is performed by the testing method of 7.3 6. construction 6.1 general in construction the construction of an apparatus for estimation shall conform to the following respective items. the shape shall be regular, and the assembly and finish of each part shall be good and strong. there is no fear for risks under an ordinary operation state, and the apparatus for estimation shall operate safely and smoothly. each part shall be so constructed as not to cause mechanical and electrical troubles and not to generate risks. the apparatus for estimation shall be so constructed not to cause the disturb of the operation of the apparatus for estimation due to water leak, water splash, dew condensation, etc. the apparatus for estimation shall be so constructed as to operate easily in maintenance and inspection and to be free from dangers. copyright japanese standards association provided by ihs under license with jsalicensee=ihs employees/1111111001, user=wing, bernie not for resale, 03/12/2007 02:52:31 mdtno reproduction or networking permitted without license from ihs -,-,- 3 k 3602-1990 6.2 structure part, a supply part of buffer solution or the like, a discharge part, and an indication recording part. fig. 1. the apparatus for estimation shall be composed of a detection structure of apparatus for estimation (an example) i ! detection part a : microbial sensor 8 : measuring ce11 - : flow of sample - : signal 6.3 detection part a detection part shall keep a microbial sensor and sample solution under a state of being in contact with each other, and shall supply a signal to be generated to an indication part stably, which is composed of the microbial sensor, a measuring cell, etc. (1) microbial sensor a microbial sensor shall be composed of an immobilized microbial membrane (the immobilized membrane of trichosporon cutaneum ifo-10466) with oxygen electrode, and the immobilized microbial membrane shall be so constructed as to be easily interchangeable. (1) sample solution feeder a sample solution feeder sends some preliminarily prepared samples to a detection part following the determined order to be measured, it should preferably have a function capable of calibrating a microbial sensor by sending the bods calibrating solution of 7.1 (5) on occasion. (2) output current recorder record the output current of a microbial sensor. an output current recorder shall be able to 7. tests 7.1 reagents reagents to be used for tests shall be as follows. water water for analysis specified in jis k 0050. glucose l-glutamic acid glucose specified in jis k 8824. l-glutamic acid specified in jis k 9047. bods calibration stock solution bods calibration stock solution is prepared by dissolving 150 mg of glucose and 150 mg of l-glutamic acid, with water in a 1000 ml volumetric flask, to the mark line. value of this calibration stock solution in accordance with the measuring method for biochemical oxygen demand (bod) specified in 21. of jis k 0102. shall be confirmed to be 220 f 10 mg/. this calibration stock solution shall be used as soon as possible after preparation. the bod bods calibration solution weighing out the bods calibration stock solution so as to have the bod value described in an instruction manual into a volumetric flask and adding water to the mark line. bods calibration solution is prepared by further, the solution which is prepared in accordance with this method and is supplied by the manufacturer may be taken as the bods calibra tion solution. buffer solution buffer solution described in an instruction manual. 7.2 testing conditions the testing conditions shall be as follows. (1) measuring cell temperature a measuring cell temperature described in an instruction manual. (1) items concerning use method selection of setting position and use ambient temperature range (ambient temperature range guaranteed by manufacturer) outline of the construction, wiring, and tubing of an apparatus for estimation. the preparation and reservation method of reagents, the reservation method of immobilized microbial membrane (when all of reagents or the like are supplied by manufacturers; the preparation method may be not described), and sanitary consideration concerning treating an immobilized micobial membrane, etc. items concerning: measurement activation of an immobilized microbial membrane preparation of measurement method and frequency of calibration measuring method treatment when measurement is stopped items concerning measurement such as the range wherein the linearity of an apparatus for estimation is effected and the considerable points on interfering substance, etc. (2) items concerning maintenance (a) standard frequency of exchange of immobilized microbial membrane or microbial sensor. (b) exchange of immobilized microbial membrane and assembly method of microbial sensor, or exchange method of microbial sensor.
