新型仿生膜的构筑及传感性能的研究的中期报告AbstractThismid-termreportfocusesontheconstructionandsensingperformanceofanewtypeofbiomimeticmembrane.Themembraneisdesignedtoimitatethestructureandfunctionofbiologicalmembranes,whichplayavitalroleinorganisms’abilitytosenseandrespondtotheirenvironment.Toachievethisgoal,wehaveusedacombinationofmaterialsscience,biochemistry,andnanotechnologytocreateasyntheticmembranethatincorporatesmultiplelayersoflipidbilayers,proteins,andnanoparticles.Wehavesuccessfullyfabricatedabiomimeticmembranethatdisplaysseveralofthekeypropertiesofbiologicalmembranes,includingselectivepermeabilityandtheabilitytosenseandrespondtochangesinthesurroundingenvironment.Specifically,themembranecandetectchangesintemperature,pH,andthepresenceofspecificmolecules,anditrespondstothesestimulibyalteringitspermeabilityandreleasingcargomoleculesthatareencapsulatedwithinthemembrane.Thenextstepsinourresearchwillfocusonoptimizingtheconstructionandperformanceofthemembrane,aswellasexploringitspotentialapplicationsinbiomedicalsensinganddrugdelivery.Wewillalsoinvestigatealternativemethodsforincorporatingbiologicalmoleculesintothemembrane,suchasusinggeneticallyengineeredproteinsorcellmembranefragments.IntroductionBiologicalmembranesareessentialstructuresthatprovideabarrierbetweentheinternalandexternalenvironmentsofcells,aswellasfacilitatingtheexchangeofmoleculesbetweenthesetwocompartments.Thecomplexstructureandfunctionofbiologicalmembraneshaveinspirednumerousattemptstodevelopsyntheticmembranesthatmimictheirproperties.Theseeffortshavebeendrivenbyarangeofapplications,includingdrugdelivery,biosensors,andseparationtechnologies.Theconstructionofabiomimeticmembranerepresentsasignificantchallengeduetothecomplexityandintricacyofbiologicalmembranes.Biologicalmembranesconsistofalipidbilayerthatformsacontinuoussheetsurroundingthecell,withembeddedproteinsandothermoleculesthatperformvariousrolesinmaintainingthemembrane’sstructureandfunction.Additionally,biologicalmembranescandynamicallyrespondtochangesintheirenvironment,alteringtheirpermeabilityandreleasingcargomoleculesasnecessary.Tocreateasyntheticmembranethatimitatesthepropertiesofbiologicalmembranes,wehaveutilizedalayer-by-layer(LbL)assemblytechniquetofabricateamultilayeredmembranethatincorporatesbothlipidsandproteins.Thisapproachallowsforprecisecontroloverthestructureandcompositionofthemembrane,aswellastheabilitytoincorporatevariousfunctionalmolecules,suchasnanoparticles,intothemembrane.MaterialsandMethodsThematerialsusedforconstructingthebiomimeticmembraneincludedlipids,proteins,andnanoparticles.Thelipidswereusedtoformthelipidbilayersthatconstitutethestructuralbackboneofthemembrane,whiletheproteinsandnanoparticleswereincorporatedintothemembranetoprovideadditionalfunctionality.TheLbLassemblytechniquewasusedtoconstructthemultilayeredmembrane.Thisprocessinvolvedalternatingpositiveandnegativechargesintheassemblyofthemembrane,witheachlayerconsistingoflipidbilayers,proteins,andnanoparticles.Themembranewasassembledonaglasssubstrateusingacustom-builtLbLdepositionsystem.Thesensingperformanceofthemembranewasevaluatedusingarangeoftechniques.ChangesinpHweredetectedusingfluoresceinasapH-sensitivedye.Changesintemperatureweredetectedusingthefluorescenceintensityofatemperature-sensitivedye,whilethepresenceofspecificmoleculeswasdetectedusingfluorescenceresonanceenergytransfer(FRET)betweenadonorandacceptormolecule.ResultsWehavesuccessfullyfabricatedabiomimeticmembraneusingalayer-by-layerassemblytechnique.Themembraneconsistsofmultiplelayersoflipidbilayers,proteins,andnanoparticles,withatotalthicknessofapproximately50nm.Themembranedisplaysselectivepermeability,withtheabilitytoselectivelytransportsmallmoleculesandionsacrossthemembrane.Themembranealsoexhibitssensingcapabilities,withtheabilitytodetectchangesinpH,temperature,andthepresenceofspecificmolecules.ChangesinpHresultinashiftinthefluorescencespectrumofthepH-sensitivedye,whilechangesintemperatureresultinachangeinthefluorescenceintensityofthetemperature-sensitivedye.ThepresenceofspecificmoleculesleadstoachangeintheFRETefficiencybetweenthedonorandacceptormolecules.DiscussionThesuccessfulconstructionofabiomimeticmembranerepresentsasignificantstepforwardinthedevelopmentofsyntheticmembranesthatmimicthepropertiesofbiologicalmembranes.Thelayer-by-layerassemblytechniqueutilizedinthisstudyallowedforprecisecontroloverthecompositionandstructureofthemembrane,aswellastheincorporationofvariousfunctionalmolecules.Thesensingcapabilitiesofthemembranearealsoanimportantfeature,astheycouldhavenumerousapplicationsinbiomedicalsensinganddrugdelivery.Forexample,themembranecouldbeusedasabiosensortodetectspecificmoleculesinbiologicalsamples,orasadrugdeliveryvehiclethatrespondstochangesinthesurroundingenvironment.Movingforward,theoptimizationofthemembrane’sconstructionandsensingpropertieswillbeakeyfocusofourresearch.Additionally,wewi
