Not for use in diagnostic procedures.vDescription of the System About the KitSystem Summary The PureLink HiPure Plasmid Purification Kits allow you to isolate high yields of highly pure plasmid DNA. The kitsare designed to efficiently isolate plasmid DNA from E. coli in 1.52 hours using a

16、nion-exchange columns, without theuse of any organic solvents or cesium chloride (CsCl. Theisolated plasmid DNA is of high purity, equivalent to twopasses through CsCl gradients, and contains low endotoxinlevels (page 19.The PureLink HiPure Plasmid DNA Purification Kits areavailable in three formats

17、 that allow you to purify plasmid DNA using different starting culture volumes (page 5.The HiPure Technology The HiPure technology is based on anion-exchange chromatography. The technology uses a patented resin composed of small particles with a uniform pore size, to provide high yields and reproduc

18、ible performance. The spacer arm with increased lengthprovides improved DNA bindingefficiency. The unique patented ion-exchange moiety provides highefficiency for separation of DNA fromcellular contaminants including RNA.Continued on next pageSystem Overview The PureLink HiPure Plasmid DNA Purificat

19、ion Kits use a patented anion-exchange resin to purify plasmid DNA to a level equivalent to two passes through CsCl gradients.The patented resin provides excellent capacity with fast flowrates, high resolution, high yield, and efficient endotoxinremoval.To purify plasmid DNA, you will harvest E. col

20、i cells,resuspend the cells in Resuspension Buffer (R3 withRNase A, and then lyse the cells with Lysis Buffer (L7.Then, you will add Precipitation Buffer (N3 to the lysate and clarify the lysate using a centrifuge. Next, you will pass the clear lysate through a pre-packed anion exchange column. The

21、negatively charged phosphates on the backbone of the DNA interact with the positive charges on the surface of the resin. The temperature, salt concentration, and pH of the solutions influence binding. Under moderate salt conditions, plasmid DNA remains bound to the resin while RNA, proteins, carbohy

22、drates and other impurities are washed away with Wash Buffer (W8. After washing, you will elute the plasmid DNA under high salt conditions with the Elution Buffer (E4. Last, you will desalt and concentrate the DNA with an alcohol precipitation step. You can complete the entire protocol in 1.52 hours

MethodsBefore StartingIntroduction Review the information in this section before star

28、ting.Guidelines are included for growing the overnight cell cultureand for determining the appropriate amounts of startingmaterial based on the plasmid copy number used. Some buffers in the PureLink HiPure Plasmid DNAPurification Kit contain hazardous chemicals.Always wear a laboratory coat, disposa

29、ble gloves, and eyeprotection when handling the buffers.Bacterial Cultures Grow transformed E. coli cells overnight in LB (Luria-Bertani medium with the appropriate antibiotic. Harvest the bacterial culture in transition between exponential phase and stationary phase. The culture should have a cell

30、density of 109 cells/mL or an optical density of 2.0 at 600 nm (OD600.Plasmid Type and Copy Number Use PureLink HiPure Plasmid DNA Purification Kits for purifying all types of plasmid DNA, including BAC (page 20, bacmids (page 24, and ssM13 DNAs (page 28.High copy number plasmids provide the best re

31、sults, with a typical yield of 26 g DNA/mL from an overnight culture. Yields from low copy number plasmids are dependent upon culture conditions and vector/host strain combinations. When isolating low copy number plasmids, use a higher volume of cell culture, as directed in the protocol. Recommended

34、btain additional buffers by ordering the PureLink HiPure BAC Buffer kit (see Additional Products , page 34. Follow the recommendations below to obtain the best results: Maintain a sterile workspace and equipment (including pipette tips and tubes to avoid DNase contamination. Ensure that no DNase is

35、introduced into the sterile solutions supplied with the kit. Use the PureLink Nucleic Acid Purification Rack for column purification (see Purification Rack , page 7. Perform all recommended wash steps for best results. Use the TE Buffer (TE provided or 10 mM Tris-HCl, pH 8.5 to resuspend the DNA pel

36、let. Using theColumnHolderThe Column Holders in the kit allow Midi and Maxi Columns to be supported in an upright position when placed in the mouth of an Erlenmeyer (or similar flask. To use the Column Holder, slip the column through the hole in the center of the Column Holder. The column with Colum

37、n Holder can then be placed in the mouth of a flask. Continued on next pagePurification Rack The PureLink Nucleic Acid Purification Rack (see page 34 is designed specifically for use with PureLink HiPure Plasmid DNA Miniprep, Midiprep, and Maxiprep Kits.The PureLink Nucleic Acid Purification Rack co

38、nsists of a Column Holder Rack (for processing 12 miniprep,8 midiprep, and 4 maxiprep columns, a Collection Tube Rack (capable of accommodating various types and sizes of recovery tubes, and a large capacity Waste Tray for collecting waste.Buffer Preparation Resuspension Buffer (R3Add RNase A to the

39、 Resuspension Buffer (R3 according to instructions on the label of the bottle. Mix well. Mark the bottle label to indicate that it contains RNase A (100 g/mL final concentration. 45、paring Cell Lysate (next page while the column is equilibrating.Continued on next pagePreparing Cell Lysate 1.For high copy number plasmids, use 13 mL of anovernight LB culture per sample in a microcentrifugetube.Note: When using 23 mL of culture, pellet 11.5 mLculture twice in the same microcentrif

46、uge tube.If you are using 5 mL of culture volume of high copyplasmids, add twice the amount of Resuspension Buffer (R3 with RNase A, Lysis Buffer (L7, and Precipitation Buffer (N3 as directed in steps 3, 4, and 5, below forbest results.For low copy number plasmids, use 1015 mL of anovernight LB cult

47、ure per sample in a 15-mL disposable tube.2.Harvest the cells by centrifuging the overnightLB culture at 4000 g for 510 minutes.Remove all medium.3.Add 0.4 mL Resuspension Buffer (R3 with RNase A tothe pellet and resuspend cells until homogeneous.Note: If cells were resuspended in a 15-mL disposable

48、tube, then transfer the cells in a microcentrifuge tube. 4.Add 0.4 mL Lysis Buffer (L7. Mix gently by invertingthe capped tube until the lysate mixture is thoroughlyhomogenous. Do not vortex. Incubate at roomtemperature for 5 minutes.Note: Do not allow lysis to proceed for more than5 minutes.5.Add 0

49、.4 mL Precipitation Buffer (N3 and miximmediately by inverting the tube until the mixture isthoroughly homogeneous. Do not vortex.6.Centrifuge the lysate at 12,000 g for 10 minutes atroom temperature.Note: If the pellet does not adhere to the bottom of thetube, incubate the tube at room temperature

50、for5 minutes to allow the lysate and gelatinous pellet toseparate. Pipet the clear lysate into another sterile tubeand centrifuge at 12,000 g for 5 minutes at roomtemperature to remove any remaining cellular debris. 7.Proceed to Binding and Washing DNA (next page.Continued on next pageBinding and Wa

51、shing DNA 1.Load the supernatant from step 6 (Preparing CellLysate onto the equilibrated column. Allow thesolution in the column to drain by gravity flow.2.Wash the column twice with 2.5 mL Wash Buffer (W8.Allow the solution in the column to drain by gravityflow after each wash. Discard the flow-thr

52、ough.3.Proceed to Eluting and Precipitating DNA, below.Eluting and Precipitating DNA 1.Place a sterile microcentrifuge tube (elution tube underthe column.2.Add 0.9 mL Elution Buffer (E4 to the column to elutethe DNA. Allow the solution to drain by gravity flow.Do not force out any remaining solution

53、.The elution tube contains the purified DNA.Discard the column.3.Add 0.63 mL isopropanol to the elution tube. Mix well.4.Centrifuge the elution tube at 12,000 g for 30 minutesat 4C. Carefully remove and discard the supernatant.5.Resuspend the DNA pellet in 1 mL 70% ethanol.6.Centrifuge the tube at 1

54、2,000 g for 5 minutes at 4C.Carefully remove and discard the supernatant.7.Air-dry the pellet for 10 minutes.8.Resuspend the DNA pellet in 50 L TE Buffer (TE.Note: Occasionally, insoluble particles may be present.These particles do not influence the quality of the DNA and can be easily removed. To r

55、emove insolubleparticles, centrifuge the DNA solution at high speed for1 minute at room temperature. Transfer the supernatant(DNA sample into a fresh tube.Storing DNA To avoid repeated freezing and thawing of DNA, store thepurified DNA at 4C for immediate use or aliquot the DNAand store at 20C for l
