AlargefractionofTCMingredientsarederivedfromplants,whosetherapeuticeffectsarelargelyassociatedwithsecondarymetabolitessuchasalkaloids,flavonoids,andsaponins.Plantsareremarkablechemists–theycan’tescapefromdangerorenvironmentalstressors,sotheyhavenochoicebuttosynthesizetheirwaytosurvival.Theirstrategiesincludesecretingbitterortoxiccompoundstodeterconsumersinthewild.Someofthesebioactiveordefensivecompoundslaterprovedpotentenoughtobenefithumanhealth,becomingthebasisofmanytraditionalandmodernmedicines.3

Foraslongashumanshavebeenhunter-gatherers,ourancestorshaveforagedforplantstoseekrelieffromtheirillnesses.Theaccumulatedwisdomfromthoseearlyexplorations,preservedinclassicalTCMtexts,hasbecomeatreasuretroveofinspirationformoderndrugdiscovery.Bydrawingontheseage-oldprescriptions,scientistshaveachievedmedicalbreakthroughsofglobalimpact,amongwhichthediscoveryofartemisinin–anantimalarialdrugextractedfromQinghao(ArtemisiaeAnnuaeHerba)–standsoutmost.Itsdiscoveryissomewhatofalegend:In1969,aChinesescientistnamedTuYouyoubegancombingthroughtheyellowedpagesofancientmedicaltomesforacureformalaria.Atthattime,outbreaksofmalariaweresweepingthroughsouthernChinaanditsneighbors.Tocurbtherampantspreadofthedisease,theChinesegovernmentlaunchedProject523.Astheoperation’sleader,TudiscoveredfrequentmentionsofQinghaoinprescriptionsfortreatingmalaria.Herteamisolatedtheactiveingredientfromtheherbanddevelopedalow-temperatureextractiontechniquethatpreservedthemolecule’sbioactivity.Tuevenvolunteeredasoneofthefirsthumantestsubjectsforthenewdrug.Herdiscoverysavedmillionsoflivesandearnedherashareofthe2015NobelPrizeinPhysiologyorMedicine.4

IdentifyingtheactiveagentinTCMprescriptions,especiallyincompoundformulas,ischallenging.ATCMdecoctionmaycontainover10plantingredients,eachcontributinghundredsoreventhousandsofmetabolites.5Thisco-existenceofnumerousmetabolitesresultingfrommulti-herbcombinationsposesatechnicalchallengeinidentifyingactiveagents.Buttheapparentcomplexityoftheseformulasembodiesadeliberatetherapeuticdesign–ratherthanseekingasingleactivesubstance,TCMemphasizesthedynamicinteractionamongmultipleingredientsthatworktogethertorestorebalanceandharmonywithinthebody.Inthissense,thedifficultyofisolatingasingleactiveagenthighlightshowTCMdiffersfundamentallyfromthereductionistapproachinconventionalpharmacology.6
