解毒药、通络药及生津药配伍对干燥综合征大鼠AQP5表达的对比分析干燥综合征诊断标准 摘要 目的 探讨解毒药、通络药及生津药配伍对干燥综合征大鼠AQP5的影响。 方法 将70只雌性SD大鼠随机分为7组,即解毒通络生津组(A)、解毒组(B)、通络组(C)、生津组(D)、强的松组(E)、生理盐水组(F)及正常对照组(G)。除正常对照组外,每只大鼠均经过造模,建立SS模型,正常对照组注射相同剂量生理盐水。各组分别给予解毒通络生津中药、清热解毒中药、活血通络中药、养阴生津中药、强的松、生理盐水治疗28 d,正常对照组不做处理。观察大鼠饮水量的变化,采用逆转录聚合酶链式反应(RT-PCR)对颌下腺的AQP5进行半定量分析。 结果 解毒通络生津组大鼠饮水量明显减少,与其他各中药组及强的松组比较差异有统计学意义(P 0.05),通过RT-PCR半定量分析显示,解毒通络生津组AQP5的表达优于其他各组(P 0.05);生津组及强的松组AQP5的表达优于解毒组和通络组(P 0.05)。 结论 生津药、解毒药及通络药配伍可改善大鼠口干症状,其机制可能与上调大鼠颌下腺AQP5的表达有关。 关键词 干燥综合征;AQP5;生津药;解毒药;通络药 R285.5 A 1673-7210(xx)07(a)-0024-03 Analysis of the expression of AQP5 in rats with Sjogren syndrome intervened by Detoxicating Medicine, Meridians-relieving Medicine and Engender Fluid Medicine WANG Dan1 ZHAO Hao2 LU Qiuju1 XUE Luan1 1.Department of Rheumatism, Yueyang Hospital of Intergrated Chinese and Western Medicine, Shanghai University of Traditional Chinese Medicine, Shanghai xx37, China; 2.Department of Rheumatism, Guanghua Hospital of Shanghai City, Shanghai 200052, China Abstract Objective To investigate the effect of Detoxicating Medicine, Meridians-relieving Medicine and Engender Fluid Medicine on AQP5 in rats with Sjogren syndrome. Methods Seventy female SD rats were randomly divided into Detoxicating, Meridians-relieving and Engender Fluid group(A), Detoxicating group (B), Meridians-relieving group (C), Engender Fluid group (D), Prednisone group (E), physiological saline group (F) and normal control group (G). The rats were made into the model of Sjogren syndrome except the rats in normal control group which were injected the same dose physiological saline. Detoxicating, Meridians-relieving and Engender Fluid Medicine, Heat-clearing and Detoxicating Medicine, Blood-activating and Meridians-relieving Medicine, Yin-tonifying and Engender Fluid Medicine, Prednisone and physiological saline were given to each group for 28 days. Nothing was given to the normal control group. The drinking changes of rats were observed, and the changes of AQP5 in submandibular gland using semiquantitative analysis of RT-PCR. Results The drinking of rats in Detoxicating, Meridians-relieving and Engender Fluid group were reduced significantly than that of the other groups (P 0.05). The expression of AQP5 in Detoxicating, Meridians-relieving and Engender Fluid group were more significance than that of the other groups through the method of RT-PCR (P 0.05). The expression of AQP5 in the Engender Fluid group and Prednisone group were more significance than that of Detoxicating group and Meridians-relieving group (P 0.05).
