1、丙泊酚联合骨髓间充质干细胞移植脊髓损伤大鼠:改善后肢电生理功能作用优于单独用药来自中国河北医科大学附属唐山工人医院王跃新所在科研团队一项最新的研究显示,丙泊酚联合骨髓间充质干细胞移植治疗可有效改善脊髓损伤大鼠后肢电生理功能,促进运动功能的恢复,起到神经保护作用。骨髓间充质干细胞在一定条件诱导下具有自我更新、多向分化、迁移的能力,可在体外能够分化培养扩增为多种遗传稳定的组织和细胞,可作为理想的神经元移植供体。移植的骨髓间充质干细胞能在宿主体内受损伤脊髓部位存活,产生并释放趋化因子向损伤区域迁移、聚集,分泌多种生长因子,抑制炎症因子的表达,诱导损伤区域微血管的再生,减轻局部的继发性炎症反应,分化成
2、神经元和胶质细胞,达到保护并促进神经元的再生重构、治疗脊髓损伤的目的。但目前的显示单一的骨髓间充质干细胞移植对神经系统损伤的修复作用仍不理想。因此王跃新等拟研究神经系统损伤后起神经保护作用的丙泊酚与骨髓间充质干细胞联合干预对脊髓损伤的治疗效果。王跃新等以打击法建立脊髓损伤大鼠模型后,分别以骨髓间充质干细胞移植和/或丙泊酚干预方法进行治疗。结果发现经骨髓间充质干细胞移植和/或丙泊酚治疗4周后,脊髓损伤大鼠脊髓组织缺失及脊髓空洞明显缩小,脊髓组织中PKH-26阳性细胞和辣根过氧化物酶阳性神经纤维数量明显增加,下肢感觉诱发电位及运动诱发电位的潜伏期明显缩短,波幅增加,下肢运动功能明显改善,且两者联合

8、n Neural Regeneration Research (Vol. 10, No. 4, 2015), propofol injection combined with bone marrow mesenchymal stem cell (BMSCs) transplantation can effectively improve hindlimb electrophysiological function, promote the recovery of motor function, and play a neuroprotective role in spinal cord inj
9、ury (SCI).Under certain conditions, BMSCs have the ability to self-renew, multi-differentiate and migrate, are able to differentiate into various tissues and cells in vitro, and can be used as ideal donors for neuron transplantation. The transplanted BMSCs can survive in the host spinal cord, genera
10、te and release chemotactic factors which migrate toward and accumulate in the injured area, secrete various growth factors, inhibit the expression of inflammatory factors, induce the regeneration of microvessels in the injured area, alleviate local secondary inflammatory reaction, differentiate into
11、 neurons and glial cells, and protect and promote neuronal regeneration, thereby achieving the purpose of treating SCI. Simple BMSCs transplantation for repair of nervous system injury is not satisfactory. Yue-xin Wang et al. established rat models of SCI using the weight drop method and treated the
12、m with BMSCs transplantation and/or propofol injection to investigate the neuroprotective effects of these two interventions. They found that at 4 weeks after BMSCs transplantation and/or propofol injection, the cavity within the spinal cord was reduced, the numbers of PKH-26-positive cells and hors
13、eradish peroxidase-positive nerve fibers apparently increased in the spinal cord, latencies of somatosensory evoked potentials and motor evoked potentials in the lower extremity were noticeably shortened, amplitude was increased and lower extremity motor function was obviously improved. Moreover, th
14、e combined effects were better than cell transplantation or propofol injection alone. The above data suggest that the transplanted HBMSCs survived in the injured spinal cord, promoted the repair of nerve fibers, and contributed to the recovery of somatosensory evoked potentials and motor evoked pote
