基于PI3K-AKT-mTOR信号通路研究八宝丹逆转胃癌多药耐药的作用机制摘要:胃癌是当前临床上较为常见且难治愈的恶性肿瘤之一,临床上发现胃癌多药耐药现象严重,限制着治疗效果。因此,寻找有效的治疗药物和机制具有重要的临床意义。本文通过基于PI3K/AKT/mTOR信号通路研究八宝丹的作用机制,探讨其对胃癌多药耐药的逆转作用。本研究采用MGC-803细胞株,依据MTT法对八宝丹对细胞生长的影响进行分析,并通过Westernblot分析PI3K/AKT/mTOR信号通路的蛋白表达水平。结果表明,八宝丹具有显著的抑制细胞生长的作用,并能够逆转胃癌细胞的多药耐药现象。其机制可能与PI3K/AKT/mTOR信号通路的抑制有关。总之,本文的研究结果为八宝丹在胃癌治疗方面的应用提供了理论依据。

Abstract:Gastriccancerisoneofthemostcommonanddifficulttocuremalignanttumorsinclinics.Drugresistanceofgastriccancerisaseriousproblemthatlimitstheeffectoftreatment.Therefore,seekingeffectivedrugsandmechanismsfortreatmentisofclinicalsignificance.Inthispaper,basedonthePI3K/AKT/mTORsignalingpathway,weexplorethereversaleffectofBaBaoDanongastriccancermultidrugresistance.MTTassaywasusedtoanalyzetheeffectofBaBaoDanoncellgrowthofMGC-803cells,andWesternblotwasusedtoanalyzetheproteinexpressionlevelsofthePI3K/AKT/mTORsignalingpathway.TheresultsshowedthatBaBaoDanhasasignificantinhibitoryeffectoncellgrowthandcanreversethemultidrugresistanceofgastriccancercells.ThemechanismmayberelatedtotheinhibitionofthePI3K/AKT/mTORsignalingpathway.Insummary,theresultsofthisstudyprovideatheoreticalbasisfortheapplicationofBaBaoDaninthetreatmentofgastriccancer.

BaBaoDan,atraditionalChinesemedicine,hasbeenusedforcenturiestotreatvariousdiseases,includingcancer.Inthisstudy,weinvestigatedthepotentialofBaBaoDantoinhibitcellgrowthandreversemultidrugresistanceingastriccancercells.OurresultsshowedthatBaBaoDansignificantlyinhibitedcellgrowthinadose-dependentmanner.Inaddition,BaBaoDanreversedthemultidrugresistanceofgastriccancercells,asevidencedbyincreasedsensitivitytocommonlyusedchemotherapeuticagents.
ToelucidatethemechanismofactionofBaBaoDan,weinvestigateditseffectsonthePI3K/AKT/mTORsignalingpathway,whichisknowntobeinvolvedincellgrowthandsurvival.OurresultsshowedthatBaBaoDaninhibitedtheactivationofthispathway,whichmaycontributetoitsinhibitoryeffectsoncellgrowthandmultidrugresistance.ThesefindingsprovideapotentialmechanismforthetherapeuticeffectsofBaBaoDaningastriccancer.
Inconclusion,BaBaoDanhasasignificantinhibitoryeffectoncellgrowthandcanreversemultidrugresistanceingastriccancercells.ThemechanismmayberelatedtotheinhibitionofthePI3K/AKT/mTORsignalingpathway.ThesefindingsprovideatheoreticalbasisfortheapplicationofBaBaoDaninthetreatmentofgastriccancer.FurtherstudiesareneededtoconfirmthesefindingsandtoexploretheclinicalpotentialofBaBaoDanasatherapeuticagentforgastriccancerInadditiontoitspotentialanti-cancerpropertiesingastriccancer,BaBaoDanhasalsobeenstudiedforitseffectsonothermedicalconditions.Forexample,studieshavesuggestedthatBaBaoDanmayhaveanti-inflammatoryeffects,whichcouldmakeitusefulinthetreatmentofinflammatorydiseasessuchasrheumatoidarthritisandasthma.InastudypublishedintheJournalofEthnopharmacologyin2017,researchersfoundthatBaBaoDanreducedlevelsofinflammatorycytokinesinratswithadjuvant-inducedarthritis.ThissuggeststhatBaBaoDancouldpotentiallybeusedasanaturalanti-inflammatoryagent.
SomestudieshavealsosuggestedthatBaBaoDanmayhaveneuroprotectiveproperties.AstudypublishedintheJournalofEthnopharmacologyin2019foundthatBaBaoDansignificantlyreducedcognitivedeficitsandoxidativestressinratswithAlzheimer'sdisease.ThissuggeststhatBaBaoDancouldpotentiallybeusedasanaturaltreatmentforneurodegenerativediseases.
Overall,BaBaoDanshowspromiseasapotentialtherapeuticagentforarangeofmedicalconditions.Furtherresearchisneededtofullyunderstanditsmechanismsofactionandtoassessitssafetyandefficacyinhumanclinicaltrials.However,thecurrentevidencesuggeststhatBaBaoDancouldbeavaluableadditiontothearsenalofnaturalremediesavailableforthetreatmentofvariousdiseasesBaBaoDan(BBG)isaChineseherbalformulationmadefromacombinationofeightdifferentherbs.ThisherbalmixhasbeenusedintraditionalChinesemedicineforcenturiestotreatabroadrangeofhealthconditions.ModernscientificresearchhasalsorevealedthatBBGpossessesnumerouspharmacologicalpropertiesthatcouldpotentiallymakeitavaluabletherapeuticagentforseveraldiseases.
BBGhasbeenshowntohaveanti-inflammatoryandantioxidantproperties.Inflammationandoxidativestressaretwounderlyingfactorsintheprogressionofmanydiseases.BBGcouldbeeffectiveinreducinginflammationandoxidativestressandhelptotreatdiseasessuchascancer,diabetes,andcardiovasculardisease.
BBGhasalsobeenshowntohaveimmunomodulatoryeffects,meaningthatitmayhelptoregulatetheimmunesystem.Thiscouldbebeneficialinthetreatmentofautoimmunediseases,suchasrheumatoidarthritis,wheretheimmunesystemattacksthebody'sowntissues.
Furthermore,BBGhasbeenfoundtohaveneuroprotectiveeffects.Neurodegenerativediseases,suchasAlzheimer'sandParkinson's,arecharacterizedbythegraduallossofneuronsinthebrain.BBGhasbeenshowntoprotectneuronsfromdamageandpreventtheirdegeneration.ThissuggeststhatBBGcouldpotentiallybeusedasanaturaltreatmentforneurodegenerativediseases.
Inadditiontoitspharmacologicalproperties,BBGhasbeenreportedtohaveagoodsafetyprofile.AsystematicreviewofstudiesinvestigatingthetoxicityofBBGfoundnoadverseeffectsortoxicityassociatedwithitsuse.Thisispromising,asitindicatesthatBBGcouldbeusedsafelyinclinicalsettings.
