2、 SHR)左室肥厚(left ventricular hypertrophy, LVH)及左室心肌形态结构的影响。方法雄性14周龄SHR 50只,随机分为复方高剂量组(SHRY1)、复方中剂量组(SHRY2)、复方低剂量组(SHRY3)、模型对照组(SHRC)、硝苯地平组(SHRX)。监测尾动脉收缩压(systolic blood pressure, SBP),每4周1次,8周后取材。测算心脏重量指数(heart weight index, HWI)、左室质量指数(left ventricular mass index, LVMI);光镜、透射电镜观察左室心肌细胞形态结构的改变。结果(1)与S

3、HRC组比较,灌胃4,8周后,各治疗组SBP显著降低(P0.01);(2)与SHRC组比较,灌胃8周后,SHRY1、SHRY2及SHRX组HWI、LVMI均明显降低(P0.05或P0.01);(3)光镜观察发现:SHRC组心肌纤维增粗或萎缩、断裂或排列紊乱,部分心肌细胞核肥大或固缩;透射电镜观察发现:SHRC组可见部分心肌细胞核膜皱缩,核染色质凝聚、趋边,偶见核固缩,部分心肌细胞线粒体明显肿胀。与SHRC组比较,SHRY1组上述病变明显减轻;SHRY2、SHRX组病变有所改善;SHRY3组病变改善不明显。结论 益气活血复方、硝苯地平均能降低SBP、HWI、LVMI,减缓SHR左室肥厚形成、发展

4、过程中左室组织结构的破坏。关键词:益气活血复方;  自发性高血压大鼠;   左室肥厚;  形态结构Effects of Yiqihuoxue Decoction on the Morphological Structure of Myocardial Cells in Left Ventricle of Spontaneously Hypertensive RatsAbstract:ObjectiveTo explore the effects of Yiqihuoxue decoction on left ventricular hypertrophy

5、 and the morphological structure of myocardial cells in left ventricle of spontaneously hypertensive rats(SHR). MethodsFifty fourteenweekold male SHR were divided randomly into 5 groups: SHRY1(treated with high dose of Yiqihuoxue decoction), SHRY2(treated with moderate dose of Yiqihuoxue decoction),

6、 SHRY3(treated with low dose of Yiqihuoxue decoction), SHRC(untreated), SHRX(treated with nifedipine). The systolic blood pressure(SBP) was measured before and during treatment every four weeks. After 8 weeks of treatment, all rats were killed. Heart weight index(HWI), left ventricular mass index(LV

7、MI) were calculated. The morphological and structural changes of myocardial cells in left ventricle of SHRs were observed with light microscope and transmission electron microscope(TEM). Results(1) After 4 or 8 weeks' treatment,  compared each treatment group with SHR-C,the SBP decreased si

8、gnificantly(P0.01).  (2)After 8 weeks' treatment, compared SHRY1, SHRY2 and SHRX with SHRC respectively, both HWI and LVMI decreased significantly(P0.05 or P0.01). (3) The results under light microscope were:in SHRC, cardiac muscle fibers were swollen, decreased, disrupted or disorganized a

9、rrangement, as well as some hypertrophic nuclear or pyknosis of myocardial cells. The results under TEM were: in some left ventricular tissues obtained from SHRC group, nuclear membrane shrank and nuclear chromatin condensed and congregated under nuclear membrane like speckle, abutting against the n

10、uclear envelope; and pyknosis was observed by chance. Mitochondria of some myocardial cells were markedly swollen with some disorganized or disrupted mitochondrial cristae which displayed like uneven vacuole. Compared with SHRC,the above-mentioned pathological changes of SHRY1 were less obvious; the

11、 pathological changes ameliorated slightly in SHRY2 and SHRX; but there was no significant improvement in SHRY3. ConclusionYiqihuoxue decoction and nifedipine can reduce the SBP, HWI, LVMI and slow down the damages of left ventricle in SHRs during the process of LVH. Key words:Yiqihuoxue decoction;&

12、#160; Spontaneously hypertensive rats;  Left ventricular hypertrophy;  Morphological structure     左室肥厚(LVH)是心肌细胞肥大所致的心脏扩大及左室重量增加,多发于高血压病中晚期,其机理目前尚未完全阐明。中医学认为,高血压LVH的可能机制在于久病气虚血淤,本虚标实。据此,本研究选择益气活血法作为防治LVH的主要治法,自拟益气活血复方,旨在探讨不同剂量益气活血复方对自发性高血压大鼠LVH及左室心肌形态结构的影响。1  材料与方法1.1&

13、#160; 药物益气活血复方由人参、黄芪、当归、川芎、赤芍、丹参、葛根、降香等组成(广州二天堂医药公司提供),加水煎成200 浓度(即1 ml药液含2 g生药)。硝苯地平,10 mg/片(广东华南制药厂生产,批号:040602)。1.2  动物及分组  清洁级雄性14周龄SHR大鼠50只,体重(257±12) g,购自上海斯莱克实验动物有限责任公司。普通饲料喂养,避免噪音及其他干扰,观察期为8周。SHR随机分为5组,即复方高剂量组(SHRY1)、复方中剂量组(SHRY2)、复方低剂量组(SHRY3)、模型对照组(SHRC)、硝苯地平组(SHRX),每组10只。按体表面积法1换算:大鼠复方每日用量为13 g/kg体重,硝苯地平每日用量为5.4 mg/kg体重。以复方换算剂量为SHRY2组给药剂量,SHRY1、SHRY3组分别为其3、1/3倍;SHRX组按大鼠换算剂量灌胃。即:SHRY1、SHRY2、SHRY3、SHRX组分别予以39 ,13,4.3 ,5.4 mg/kgd药物灌胃;SHRC组予等体积蒸馏水灌胃。动物禁食12 h后灌胃,1次/d。将药物按不同剂量以蒸馏水稀释,最终每只动物均予5 ml液体灌胃。干预8周间,每周称重1次,根据体重调整药量。1.3  指标检测1.4  统计学处理所有计量资料均采用 ±
