1、红茶提取物茶多酚产品规格: 30%,60% 茶多酚检测方法: Folin D法有效成分: 茶多酚产品来源: 山茶科植物山茶的叶提取方法: 乙醇提取产品外观: 棕黄色粉末1. 理化性质分子结构类型:一类多酚化合物溶解性:易溶于水及乙醇,味苦涩2. 质量标准检验2.1 鉴别:(对照品:没食子酸)2.1.1 TLC检查,与对照品Rf值、颜色一致2.1.2 IR检查,与对照品指纹图谱吻合(红外吸收光谱)2.2 红茶茶多酚的检测方法:2.2.1 原理福林酚试剂氧化茶多酚中-OH基团并显蓝色,最大吸收波长为765nm, 用没食子酸作校正标准定量茶多酚。2.2.2 试剂及设备1. 色谱纯乙腈2. 甲醇1.
2、甲醇水溶液(体积比)=7:32. 碳酸钠(Na2CO3)3. 福林酚试剂4. 10%福林酚试剂(现配):将20mL福林酚试剂转移到200mL容量瓶中,用水定容并摇匀。5. 7.5% Na2CO3溶液(质量浓度):称取37.50g0.01gNa2CO3(10.3),加适量水溶解,转移至500mL容量瓶中,定容至刻度,摇匀。6. 没食子酸标准储备溶液(1000g/mL): 称取0.110g0.001g没食子酸(GA,相对分子质量188.14),于100mL容量瓶中溶解并定容至刻度,摇匀.7. 没食子酸工作液:用移液管分别移取1.0,2.0,3.0,4.0,5.0mL的没食子酸标准储备溶液(10.8
3、)于100mL容量瓶中,分别用水定容至刻度,摇匀,浓度分别为10,20,30,40,50g/mL。2.2.3 样品液配置准确称取10g红茶提取物样品,用适量水溶解,转入100mL容量瓶中,加水定容至刻度,摇匀,待测。 2.2.4 样品含量的检测用移液管分别取没食子酸工作液、水及测试液各1.0mL于刻度试管内,在每个试管内分别加入5.0mL的10%福林酚试剂,摇匀。反应3min8min内,加入4.0mL7.5% Na2CO3溶液,加水定容至刻度、摇匀。室温下放置60min。用10mm比色皿、在765nm波长条件下用分光光度计测定吸光度(A)。根据没食子酸工作液的吸光度(A)与各工作溶液的没食子酸
4、浓度,制作标准曲线。 2.2.5 结果计算比较试样和标准工作液的吸光度,按下式计算:式中:A样品测试液吸光度;V样品液体积;d稀释因子(稀释倍数);SLOPEstd 没食子酸标准曲线的斜率;m样品干物质含量,%;m1样品质量,单位为克(g)。2.3 检查:干燥失重: 2%灰分: 0.5%重金属: 百万分之五农药残留: 百万分之二溶剂残留:0.1%2.4 卫生检查细菌总数:1000/g霉菌总数:100/g大肠杆菌: 无沙门菌: 无2.5 粒度: 均匀接近细微颗粒2.6 包装: 双层朔料袋封口、外包装纸桶、桶外印有品名、毛重、批号、生产单位、生产时间、有效时间、贮运方法等。2.7 有效期: 2年2

Gallic acid working solution: accurately transfer 1.0, 2.0,3.0,4.0,5.0mL Gallic acid standard store solution into 100mL volumetric flask, and mix with water to volume. The concentration series are 10
12、, 20, 30, 40, 50g/mL.2.2.3 Preparation for test sampleAccurately taking 10g of Black Tea Extract samples dissolved in appropriate water, then transfer into 100mL volumetric flask, adding water to volume.2.2.4 Sample content testPartly taking 1.0mL Gallic acid working solution, Water, Test sample add
13、 into 3 graduated test tube. Each test tube be add into 5.0mL 10% Folin-Ciocalteu solvent, mixed. Let the liquid reaction 3min8min, then add 4.0mL 7.5% Na2CO3 solution and water to volume, mixed. Lay at room temperature. Detect the absorbance(A) by a 10mm cuvette at 765nm wave.Making the working cur
