第第页RatAntiANA/ENAIgGELISA大鼠抗核抗原试剂盒说明书RatAntiNuclearAntigens（ANA/ENA）IgGspecificELISAKitcat650230ANAINTENDEDUSETheRatANA（AntiNuclearAntigens）IgGELISAKitisanimmunoassaysuitableforquantifyingortiteringIgGantibodyactivityspecificforextractablenuclearantigens（ENA）inserumorplasma.Otherbiologicalfluids,includingtissueculturemedium,maybevalidatedforuse.Forresearchuseonly（RUO）,notfordiagnosis,cureorpreventionofthedisease.GENERALINFORMATIONAntibodiesreactivewithautologousnuclearcomponents,suchasDNAandhistones,canrepresentanautoimmunebasisforpathologicalconditionssuchassystemiclupuserythematosis（SLE）inhumans,andinmicehomozygousforthelymphoproliferationspontaneousmutation（Faslpr）,asystemicautoimmunitywithmassivelymphadenopathyassociatedwithproliferationofaberrantTcells,arthritisandimmunecomplexglomerulonephritis.TheseconditionsincludeelevatedlevelsofantidsDNAandotherantinuclearantigen（ANA）antibodieswhichoftenincreaseastheanimalages.Also,theexpandeduseinthedrugindustryofbiologicalmodifiershasbeenassociatedwithproductionofautoantibodies,ofwhichmice,andpossiblyalsootherhostssuchashumans,monkeysandrats,aresusceptible.Aprototypediseaseinmiceislupuscausedbythedrugminocycline,withelevatedantidsDNAamongotherautoantibodiesandpathologicalconditions.RecentinvestigationshavefocusedontheroleofinnateimmunemechanismsasunderlyingcauseoftheANAtypeautoimmunity；thesemaybeinducedbydrugs,includingvaccinesandadjuvants,withaging,orwithotherhealthconditions.PRINCIPLEOFTHETESTTheRatANAIgGELISAkitisbasedonthebindingofratANAIgGinsamplestoENAimmobilizedonthemicrowells,andANAIgGantibodyisdetectedbyantiRatIgGspecificantibodyconjugatedtoHRP（horseradishperoxidase）enzyme.Afterawashingstep,chromogenicsubstrate（TMB）isaddedandcolorisdevelopedbytheenzymaticreactionofHRPonthesubstrate,whichisdirectlyproportionaltotheamountofANAIgGpresentinthesample.StoppingSolutionisaddedtoterminatethereaction,andabsorbanceat450nmisthenmeasuredusinganELISAmicrowellreader.TheactivityofratIgGantibodyinsamplesiscalculatedrelativetoANAcalibrators.SpecificityPurifiedmammalianENAisusedtocoatthemicrowells；thustheassayisspecificforantibodiesdirectedtoENA.TheantiRatIg
