1、医学报告Medical ReportmicroRNA的异常表达在结直肠癌等众多恶性肿瘤的发生发展过程中扮演重要的角色。我们的前期研究证实miR-133b是一种新发现的在结直肠癌中具有抑瘤作用的microRNA，但仍有许多关键的问题没有得到解答，需进一步深入研究。本课题在前期工作的基础上，拟开展研究工作如下： 通过靶点筛选及细胞功能验证，对miR-133b在结直肠癌中的作用靶点进行系统的分析和深入的研究，有望阐明miR-133b在结直肠癌中的作用机理；2）通过在大样本的结直肠癌标本中检测miR-133b及其靶基因的表达特征，分析miR-133b与结直肠癌各病理指标及临床指标之间的关系；3） 从遗

2、传学和表观遗传学角度分析了miR-133b在结直肠癌中表达失调的可能原因。通过本研究有望阐明miR-133b在结直肠癌中的作用机制，更为结直肠癌的基因治疗提供新的思路与实验线索，具有重要的理论和实际意义。The dysregulation of microRNAs is an important step in the development of many cancers, including colorectal cancer (CRC). Our initial research confirms that miR-133b is a newly discovered microRNA

3、which could suppress the growth of tumor for CRC however, there are still lot of key issues without solution which need our further research. We plan to do research on the issue on the basis of our previous work achievement as follows:We will make systemic analysis and further research on the effect

4、ed targets of the miR-133b in the CRC by target screening and cell function verification, and its expected to explain the mechanism of the miR-133b in the CRC.We plan to find out the features of the miR-133b as well as its target genes by examining the large CRC samples, and analyse the relationship

5、 between pathological index together with the clinical index and miR-133b.We will analyse the possible causes of the dysregulation in the CRC for miR-133b from the aspects of HYPERLINK app:ds:genetics Genetics and Epigenetics. Its expected to explain the mechanism of the miR-133b in the CRC, and provide the CRC gene therapy with new methods
